    Steve,

if you are buying the Canon EF 100mm f/2.8 macro USM the autofocus is quite reliable, but in any case I would go for the manual focus port if the price difference is not too big .... I think you can use a ReefNeet external diopter with the proper adapter ring or a Canon 250D internal diopter.

With the focus port on ikelite there is NO space for a diopter (not even a UV filter!) Have been there, tried that and scratched the port (wasn't even mine!!!) I don't know about the auto focus though. On the plus side if you plan to use external diopter it does position the lens very close to the port which I believe is important when using external diopters.

  4. You could choose the modular system. Different extension rings will permit you to use diopters or be without them. You can then also add extenders. However, it is somewhat more expensive.

  5. The standard 5505.45 port will safely fit a single element diopter with my 7D housing (just doublechecked with a +4), I agree it wont fit a 250D, the difference is that small. Make sure you put the port on last if you do this.

If you really want the 250D, you could just get the next port size up, its only .4 inch/1cm different which will allow room and should make negligible difference to standoff difference or the like. The 5505.45 is for 5.1 inch lenses, the 5505.50 port is for 5.5 inch lenses.

I use AF only, if I want to use 'manual', I just turn off AF on the shutter button and use the AF-On button on the back to roughly focus to the distance I need then move the housing to try and improve the manual focus if needed - I usually cant keep still enough in water at that magnification to use manual focus much anyhow.

I think the standard port manual focus version for Ikelite might be quite a pain to use with a subsee adapter as the knob is forward of the port rather than reversed if not even potentially unusable going by the pictures, but someone else may be in a better position to comment.
